Jodi Lyn O'Keefe was born on October 10, 1978, in Cliffwood Beach, New Jersey, United States, to Jack O’Keefe and Noreen O’Keefe. She grew up in New Jersey alongside her two older sisters. Inspired by her eldest sister, Heather, she began modeling at the young age of eight. O'Keefe worked as a model for several notable brands, including DreamPhone and Gitano Jeans.
She attended Saint John Vianney High School but left before graduating to pursue a full-time career in acting.
O'Keefe made her acting debut in 1995, portraying Maggie Cory on the soap opera Another World. Her breakout role came shortly after in 1996, when she was cast as Cassidy Bridges on the popular television series Nash Bridges.
She gained further recognition for her portrayal of Gretchen Morgan in the hit series Prison Break (2007–2009) and also appeared as Mikayla in an episode of The Big Bang Theory in 2009. Her role as Josette Laughlin in the supernatural series The Vampire Diaries in 2014 added to her growing television repertoire.

She also starred as Lionel Davenport in Hit the Floor (2014) and appeared as Ronnie Hillman in Lucifer (2016).
O'Keefe made her film debut in 1998, playing the role of Sarah Wainthrope in Halloween H20: 20 Years Later. She gained widespread popularity for her role as Taylor Vaughan in the 1999 teen romantic comedy She’s All That.
Her subsequent film work includes roles in The Crow: Salvation (2000) as Lauren Randall, Whatever It Takes (2000) as Ashley Grant, Devil in the Flesh 2 (2000) as Debbie Strong, Falling in Love in Pongo Ponga (2002) as Shawnee, and Red Rover (2003) as Kylie Logan. Additional credits include Out for Blood (2004), Mummy an’ the Armadillo (2004), Venice Underground (2005), American Identity (2007), The Frozen Ground (2013), and Merry ExMas (2014), in which she played Ashley Williams.
As of recent reports, Jodi Lyn O'Keefe is not married and has no children. She was previously in a relationship with actor John Cusack from 2003 to 2009. Earlier in her career, she dated Al Santos in 1995, Don Johnson in 1997, and Jamie Gomez in 1998.
