Lee began his career performing stand-up comedy at various venues in London before transitioning to television. He gained national attention as a co-presenter of Channel 4's satirical news and comedy program, The 11 O’Clock Show. Alongside Mackenzie Crook and Daisy Donovan, Lee hosted the show, which also featured rising comedic talents such as Ricky Gervais and Sacha Baron Cohen, performing as his character Ali G.

One of the defining moments of Lee’s radio career occurred in December 2018, when he helped save the life of a suicidal listener who had called into his show. The caller had taken an overdose and was lying on the street in Plymouth when he reached out to Lee. By keeping the man on the line for over 30 minutes, Iain assisted emergency services in locating him, ultimately helping to save his life. The act was widely praised and brought attention to the power of live broadcasting in mental health support.
